Phage-assisted evolution yields compact, efficient prime editors (PE6a-g)

Broad Institute researchers used phage-assisted evolution to improve the editing efficiency of compact reverse transcriptases by up to 22-fold and generated prime editors 516-810 base pairs smaller than PEmax. The resulting PE6a-g editors enhanced therapeutically relevant editing in patient-derived fibroblasts and primary human T-cells, and achieved 40% loxP insertion in mouse cortex after dual-AAV delivery.
